作用機(jī)理
OKT3單克隆抗體特異性地與CD3的ε鏈/T淋巴細(xì)胞抗原受體復(fù)合物發(fā)生反應(yīng)。CD3的復(fù)合物包括 γ、 δ和ε鏈，它是TCR復(fù)合物的一部分，由成熟的T淋巴細(xì)胞和胸腺細(xì)胞譜系表達(dá)。CD3通過信號(hào)轉(zhuǎn)導(dǎo)增強(qiáng)抗原識(shí)別。而OKT3抗體是一種免疫抑制劑，在肝臟、心臟和腎臟同種異體移植排斥方面，這已被證明是一種有效的治療劑。
Description
The OKT3 monoclonal antibody specifically reacts with the ε chain of the CD3/T lymphocyte antigen receptor complex. The CD3 complex contains γ, δ, and ε chains, and it is part of the TCR complex, expressed by all mature T lymphocytes and by the thymocyte lineage. CD3 enhances the antigen recognition by signal transduction.
The OKT3 antibody is an immunosuppressive, which has proven to be an effective therapeutic agent in liver, heart, and renal allograft rejection.

CD3單克隆抗體（OKT3）/抗人CD3抗體是由肽鏈以非共價(jià)鍵組成的復(fù)合分子，在成熟T細(xì)胞表面表達(dá)，對(duì)阻斷急性同種異體排斥反應(yīng)起主要作用。 CD3分子與TCR組成復(fù)合受體分子，激活酪氨酸激酶，促使CD3分子免疫受體酪氨酸活化基序中的酪氨酸（Y）磷酸化。磷酸化的酪氨酸（pY）進(jìn)一步磷酸 化下游含酪氨酸的蛋白，從而引起激酶活化的級(jí)聯(lián)反應(yīng)，調(diào)控T細(xì)胞增殖和活化的靶基因，引起基因的表達(dá)和轉(zhuǎn)錄，T細(xì)胞由靜止?fàn)顟B(tài)轉(zhuǎn)為增殖和活化狀態(tài)。
CD3分子具有穩(wěn)定TCR結(jié)構(gòu)和傳遞活化信號(hào)的作用。針對(duì)CD3分子的單克隆抗體能夠激發(fā)或阻斷T細(xì)胞活化信號(hào)轉(zhuǎn)導(dǎo)，清除效應(yīng)T細(xì)胞或誘導(dǎo)調(diào)節(jié)T細(xì)胞的產(chǎn)生。
